Improvements of protein properties have first been performed by using classical protein engineering techniques based on rational design and structural approaches. However, directed evolution technologies have an increasing impact in technological platforms in order to address the need for Industrial protein optimization
Directed evolution is a powerful approach for improving any screenable property of a protein.
There are three basic steps in the process of directed evolution :
- creating a library of genetic diversity encoding new variants of the native protein ; Protéus has developed two revolutionary technologies for generating new diversity, L-Shuffling™ & EvoSight™
- expressing these genes to produce new functional proteins
- screening these new proteins to identify the ones displaying the appropriate characteristics.

Genes coding for new proteins with improved characteristics can then be used as parental variant genes in a series of subsequent rounds of directed evolution, leading rapidly to optimized proteins.
